● 產(chǎn)品簡(jiǎn)介:

本產(chǎn)品是由11條帶狀雙鏈DNA條帶組成,適用于瓊脂糖凝膠電泳中DNA條帶的分析。

    本產(chǎn)品為即用型產(chǎn)品,已含有1×loading buffer,可根據(jù)實(shí)驗(yàn)需要,直接取2-5μl電泳,使用方便,電泳圖像清晰。本產(chǎn)品中11條帶分為50100,150200,250,300,350400, 500, 6001000bp,其中300bp條帶約為100ng/5μl,其余條帶濃度大約50ng/5μl。


● 儲(chǔ)存條件:

  4 貯存6個(gè)月,-20貯存 1年。

● 使用方法:

1. 2-5μl本產(chǎn)品加入到瓊脂糖凝膠的加樣孔中(每1mm×1mm加樣孔加1μl,如果加樣孔寬于

5mm,可以適當(dāng)增加上樣量)就行電泳。

2. 建議電泳條件:凝膠濃度為2.0-2.5%,凝膠長(zhǎng)度6-7cm,電泳電壓4-10v/cm,電泳時(shí)間45-50分鐘。

3. 通過EB染色后紫外燈下觀察條帶。

注:1 如果凝膠中預(yù)先加入EB,電泳結(jié)束后紫外燈下觀察,200bp以下條帶亮度較弱,此問題可以通過凝膠后染的方法解決(即溶膠時(shí)不加EB,電泳結(jié)束后再進(jìn)行EB染色)。

2 如果使用Goldview等染料就行染色,由于靈敏度比EB低,請(qǐng)酌情增加Marker的上樣量。

● 注意事項(xiàng):

1. 瓊脂糖的質(zhì)量對(duì)DNA的電泳有很大影響,電泳時(shí)請(qǐng)盡量使用質(zhì)量?jī)?yōu)等的瓊脂糖。

2. 請(qǐng)使用新鮮配制的電泳緩沖液和新鮮配制的瓊脂糖凝膠進(jìn)行電泳,以保證Marker良好的分離效果。

3. Marker適用于DNA片段大小的確定和對(duì)DNA含量的粗略定量,不適用于DNA片段含量的精確定量。
