植物RNA提取試劑


產(chǎn)品簡介:

RNAplant試劑可從植物組織,特別是富含多酚或淀粉的植物組織中(如馬鈴薯塊莖,白松松針或嫩苗,和西紅柿葉等)提取高純度的總RNA。

100 ml可處理100 mg組織200次。

保存條件:室溫運輸,4保存,可保存6-12個月。 

準備試劑液氮,研缽,無RNase離心管,5 M NaClNC1050F 5M 氯化鈉 RNase-free,DNase-free),氯仿,異丙醇,75%乙醇(無RNase水配制),無RNase水。


小規(guī)模提取操作步驟(樣品<0.1 g ,使用小規(guī)模提取方法):

1.      取不多于0.1 g植物組織,在液氮中研碎,轉(zhuǎn)移到1.5 ml 離心管中,加0.5 ml提取試劑,振蕩至徹底混勻。

注:振蕩不要太劇烈,否則容易導致基因組DNA污染。

2.      室溫放置5分鐘。

3.      4℃ 12, 000 rpm離心2-3分鐘,上清轉(zhuǎn)入新的無RNase離心管。

4.      上清中加入0.1 ml 5 M NaClNC1050F 5M 氯化鈉 RNase-free,DNase-free),溫和混勻。

5.      加入0.3 ml 氯仿,上下顛倒混勻。

6.      4℃ 12,000 rpm離心10-15分鐘,取上層水相轉(zhuǎn)入新的無RNase離心管。

7.      加與所得水相等體積的異丙醇,混勻,室溫放置10分鐘。

8.      4℃ 12,000 rpm離心10-15分鐘。棄掉上清,注意不要倒出沉淀。加1 ml 75%乙醇沖洗沉淀。(沉淀可能很難看見,應小心操作。)

9.      4℃ 5000-7000rpm離心3分鐘。倒出液體,注意不要倒出沉淀。剩余的少量液體短暫離心,用槍頭吸出,室溫晾干1-2分鐘至沉淀干燥。

10.  加適量無RNase水充分溶解RNA,-70℃保存。

注意事項:

RNAplant有刺激性味道,操作時請在通風處進行。
